Coimmunoprecipitation (nuclear extracts)

1. Resuspend protein A Sepharose or protein G sepharose beads in IP BUFFER at 100 mg/ml

2. Swell beads by rotating 30 mins at RT. Wash 2x with IP BUFFER and resuspend at 100 mg/ml

3. For each IP reaction, mix 40 µl of 100 mg/ml protein A Sepharose with 2-20 µl antibodies in 400 µl IP BUFFER, containing protease inhibitors. Rotate over night at 4˚C.

4. Wash beads three times with IP BUFFER ( spin 2,000 rpm, 1 min and remove supernatant).

5. Transfer nuclear extract to tube containing washed antibody conjugate. Rotate at 4˚C, 2-4h.

6. Wash beads 8 times with 1 ml IP BUFFER (ice-cold) ( spin 1,000 rpm, 1 min and remove supernatant).

7. Resuspend in 10ul 6x protein sample buffer, heat for 5 min. at 95C, spin at 20 000g for 5 min. Move supernatant to new tube and use it for SDS PAGE.

IP BUFFER: 50 mM Tris-HCl pH7.5, 150 mM NaCl, 0.05% NP40.
